RESUMO

En investigaciones previas realizadas in vitro con solución saturada de sacarosa, eugenol y polietilenglicol 400 se comprobó su eficacia fungicida sobre Candida albicans.Se comparó en las mismas condiciones el efecto fungicida con nitrato de econazol. Una suspensión de Candida albicans con nitrato de econazol se repicó en Agar Sabouraud 20 a distintos tiempos para demostrar la viabilidad de la cepa.El tiempo promedio de muerte de Candida albicans en la solución saturada de sacarosa fue de 300 segundos, en nitrato de econazol fue de pocos segundos. Aunque existen diferencias significativas en el tiempo de muerte de la cepa ensayada, la solución saturada de sacarosa sería una contribución terapéutica alternativa, en aquellos pacientes con lesiones cutáneas crónicas que no responden a los tratamientos tradicionales (AU)

The purpose of this work was to determine the in vitrobioactivity of a sucrose saturated solution with eugenol (Dickinson, from Dr. Preston Lab. Argentine) and polyethylenglycol 400 ( Merck, from Merck Chemistry Argentine) over Sporothrix schenkii. This dimorphic fungi causes sporothricosis, a sub-acute or chronic infection that affects mainly skin, showing an ulcerous initial lesion with nodular lymphangitis. A strain suspension in Sabouraud dextrose 2% broth (Merck, from Merck Chemistry Argentine) equivalent to Mc Farland N degrees 5 was used, to which the sucrose saturated solution with eugenol and polyethylenglycol 400 was added. Strain viability was measured by seeding aliquots of the original suspension every 15 seconds after initial contact on Sabouraud dextrose agar plates (Britania, from Britania Lab. S.R.L. Argentine). These plates were incubated at 28 degrees C and periodically inspected during 30 days. The strain was viable until 2 min of contact with the solution, after that time no growth was detected. Fungal death may be due to cellular dehydration as a consequence of the low water activity of the referred saturated solution.

El presente estudio tuvo por objeto lograr disminuir el tiempo de sobrevida de Candida albicans incorporada a una solución sobresaturada de azúcar con eugenol y polietilenglicol 400 (PEG). El tiempo de muerte para Candida albicans en la solución sobresaturada de azúcar + eugenol se estimó en dos días. Con el agregado de PEG 400 se abrevió este tiempo a minutos. Estos resultados nos sugieren que el PEG 400 colaboraría en el descenso de la actividad agua (aw) y como consecuencia la disminución de sobrevida de Candida albicans

Joseph, A. y cols. A la solución sobresaturada de azúcar se incorporó eugenol, con el objeto de acelerar la muerte de las cepas fúngicas ensayadas. Se observaron diferencias significativas en el tiempo de muerte de los hongos estudiados con respecto a los ensayos anteriores con solución sobresaturada de azúcar

Addition of 0.3% (w/w) of potassium sorbate (a permitted food additive) coupled with a reduction of pH from 7 to 6 or 5.5, resulted in a dramatic decrease in the survival of two S. aureus strains inoculated (10(8) c.f.u./ml) in laboratory media supplemented with sugar (227 g sugar/100 g water) and incubated at 35 degrees C. These in vitro results may be of potential value to improve the efficacy of sugar therapy for the treatment of certain infected wounds; however, clinical trials are needed to confirm these findings.

Essential oil of clove, dispersed (0.4% v/v) in a concentrated sugar solution, had a marked germicidal effect against various bacteria and Candida albicans. Staphylococcus aureus (five strains), Klebsiella pneumoniae, Pseudomonas aeruginosa, Clostridium perfringens, and Escherichia coli inoculated at a level of 10(7) cfu/ml, and C. albicans (inoculum 4.0 x 10(5) cfu/ml) were killed (greater than 99.999%) after 2-7 min in a laboratory broth supplemented with 63% (v/w) of sugar, and containing 0.4% (v/w) of essential oil of clove. Added organic matter (i.e. human or bovine serum) did not impair its antimicrobial activity. Sugar was not necessary for the antimicrobial activity of clove oil, but the concentrated sugar solution provided a good vehicle for obtaining an oil dispersion that is relatively stable for certain practical applications.

This study reports the extent and character of plasmolysis and other morphological changes as shown by electron microscopy in a strain of Klebsiella pneumoniae and with sucrose or polyethylene glycol 400 (PEG-400) as the plasmolysing agent at a water activity of 0.935. Both solutes produced severe plasmolysis in K. pneumoniae cells; PEG-400 also caused some cell wall collapse and finger like extrusions to emerge from the bacterial cell.

It was found that concentrated polyethylene glycol 400 (PEG 400) solutions have significant antibacterial activity against various pathogenic bacteria, including Klebsiella pneumoniae, Pseudomonas aeruginosa, Escherichia coli, and Staphylococcus aureus. This effect might be attributed to two effects: lowering of water activity and, superimposed on this, the specific action of PEG-400 molecules on bacterial cells. Phase-contrast microscopic observations of cells placed in contact with PEG 400 revealed clumping and morphological changes of bacterial cells. The larger changes in appearance were evidenced by the species which were more rapidly killed by PEG 400. The results obtained suggested that concentrated PEG 400 solutions may have a potential value in medicine as a topical antibacterial agent. The feasibility of this application is the subject of present investigation.

RESUMO

The use of sugar for the treatment of infected wounds was investigated in in vitro experiments with bacteria pathogenic to humans, such as Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae, and Staphylococcus aureus. Studies showed that solutions of appropriate sugar concentration incubated at pH 7.0 and 35 degrees C were lethal to the bacterial species studied. On the basis of these results, it is proposed that an important function of sugar in the treatment of infected wounds is to create an environment of low water activity (aw), which inhibits or stresses bacterial growth.
